Clinical study of tesamorelin has centered on adults with HIV-associated lipodystrophy, a condition in which abdominal fat accumulates while peripheral fat is lost. In controlled trials, treated participants showed reductions in visceral adipose tissue measured by imaging, alongside modest shifts in some lipid values. Effects on subcutaneous fat were smaller and less consistent across studies. Whether these changes translate into fewer cardiovascular events remains an open question, because the trials were not designed or powered to answer it.
Tesamorelin is a synthetic peptide that acts as an analog of growth hormone-releasing hormone, a natural hypothalamic signal. Its sequence corresponds to the forty-four amino acid form of the human hormone, with a small acyl group attached near the amino terminus. That modification slows enzymatic breakdown and extends the time the peptide remains active in circulation. The compound was developed as a pharmacological way to raise endogenous growth hormone output rather than supplying the hormone directly.
Clinical interest in tesamorelin arose from the need to address visceral adiposity in people living with HIV. Antiretroviral therapy improved survival but was associated in some patients with central fat accumulation, altered lipid profiles, and metabolic complications. This condition, often called HIV-associated lipodystrophy, involves excess visceral adipose tissue that is difficult to manage through diet and exercise alone. Investigators evaluated tesamorelin because GHRH analogs can stimulate growth hormone secretion and influence fat distribution without direct liposuction or invasive procedures.
A Phase 3 program led to regulatory approval in the United States in 2010 for reduction of excess visceral abdominal fat in adults with HIV and lipodystrophy. Subsequent studies examined effects on liver fat, muscle area, and metabolic markers, with mixed findings for some endpoints. Long-term cardiovascular outcomes and effects on mortality remain uncertain because most trials were relatively short and focused on imaging-based fat measurements. Use in populations without HIV has been studied experimentally but is not part of the approved indication.
Tesamorelin is a synthetic analog of growth hormone-releasing hormone, a peptide hormone produced by the hypothalamus. The molecule retains the 44-amino-acid sequence of human GHRH and carries a trans-3-hexenoyl modification at its N-terminus. This modification increases resistance to enzymatic degradation and extends the peptide's functional stability relative to native GHRH. The compound is supplied as a lyophilized powder for reconstitution and subcutaneous administration in clinical settings. Its development code was TH9507, and it belongs to the GHRH analog class. It is not a growth hormone product; instead, it acts upstream to stimulate endogenous growth hormone release.

=== Pharmacokinetics === The metabolism of 2C-EF has been studied in vitro. It undergoes demethylation at position 2 or 5, hydroxylation, and deamination, as well as acetylation and glucuronidation. Oxidative deamination, mediated mainly by monoamine oxidase (MAO) enzymes, is the main route of metabolism for 2C-EF.
==== Carbon ==== Stable isotope analysis of carbon in human bone collagen allows bioarchaeologists to carry out dietary reconstruction and to make nutritional inferences. These chemical signatures reflect long-term dietary patterns, rather than a single meal or feast. Isotope ratios in food, especially plant food, are directly and predictably reflected in bone chemistry, allowing researchers to partially reconstruct recent diet using stable isotopes as tracers. Stable isotope analysis monitors the ratio of carbon 13 to carbon 12 (13C/12C), which is expressed as parts per thousand using delta notation (δ13C). The 13C and 12C ratio is either depleted (more negative) or enriched (more positive) relative to a standard. 12C and 13C occur in a ratio of approximately 98.9 to 1.1.
The aim is to come up with tools that can pre-screen patients with four tumour types for the MSI-H biomarker, namely endometrial, gastric, small intestinal, and biliary cancers. In January 2024, the company announced it would acquire Harpoon Therapeutics for $680 million. With this purchase, Merck expands its portfolio of oncological drugs. The main positions are HPN328, an activator of T-cells that is being researched to treat advanced cancer patients associated with DLL3 expression (delta-like ligand 3), an inherent small cell lung cancer (SCLC), neuroendocrine tumors, and several other species. Merck's portfolio will also be complemented by T-cell attractions using the patented Harpoon Tri-specific design for T cell activation (TriTAC). According to engineering protein technology, tumor cells are destroyed by the patient's own immune cells, and the ProTriTAC platform works with the TriTAC platform to develop a therapeutic agent that attracts T-cells, but is inactive until it reaches the tumor. In April 2024, Merck completed the acquisition of Abceutics for $208 million. In July 2024, Merck completed the acquisition of EyeBio for $3 billion. In October 2024, Merck announced the acquisition of Modifi Biosciences for $1.3 billion. In January 2026, the Wall Street Journal reported that Merck was in talks to acquire Revolution Medicines for ~$30 billion.
=== Protein degradation === Arginine phosphorylation by McsB kinase marks proteins for degradation by a Clp protease. The arginine phosphorylation system, which is widely distributed across Gram-positive bacteria, appears to be functionally analogous to the eukaryotic ubiquitin–proteasome system.

The other feature was a new cooling arrangement positioned aft (single ducted water and oil radiators assembly) that reduced the fuselage drag and effects on the wing. Later, after much development, they discovered that the cooling assembly could take advantage of the Meredith effect, in which heated air exited the radiator with a slight amount of jet thrust. Because NAA lacked a suitable wind tunnel to test this feature, it used the GALCIT 3.0 m (10 ft) wind tunnel at the California Institute of Technology. This led to some controversy over whether the Mustang's cooling system aerodynamics were developed by NAA's engineer Schmued or by Curtiss, as NAA had purchased the complete set of P-40 wind tunnel data and flight test reports. The NA-73X was also one of the first aircraft to have a fuselage lofted mathematically using conic sections; this resulted in smooth, low-drag surfaces. To aid production, the airframe was divided into five main sections—forward, center, rear fuselage, and two wing halves—all of which were fitted with wiring and piping before being joined. The prototype NA-73X was rolled out in September 1940, just 102 days after the order had been placed; it first flew on 26 October 1940, 149 days into the contract, an uncommonly short development period even during the war. With test pilot Vance Breese at the controls, the prototype handled well and accommodated an impressive fuel load. The aircraft's three-section, semi-monocoque fuselage was constructed entirely of 24S aluminum alloy (a type of Duralumin) to save weight.
In rats, NO inhibits the granulocyte-macrophage colony-stimulating factor (GM-CSF)-mediated maturation of dendritic cells, and in humans it inhibits the TNF-alpha-mediated maturation of human dendritic cells, through cyclic GMP-dependent mechanisms. NO prolongs the ability of human dendritic cells to internalize antigens at sites of inflammation, therefore modulating the beginning steps leading to antigen-specific immune responses. NO production has been implicated as relevant to the pathology of asthma. People with asthma show an increased expression of iNOS in airway epithelial cells and an increased level of nitric oxide in exhaled air.
=== Nuclear spins and magnets === All nucleons, that is neutrons and protons, composing any atomic nucleus, have the intrinsic quantum property of spin, an intrinsic angular momentum analogous to the classical angular momentum of a spinning sphere. The overall spin of the nucleus is determined by the spin quantum number S. If the numbers of both the protons and neutrons in a given nuclide are even then the system prefers to minimize its spin, giving S = 0, i.e. there is no overall spin. Then, just as electrons pair up in nondegenerate atomic orbitals, so do even numbers of protons or even numbers of neutrons (both of which are also spin-1/2 particles and hence fermions), giving zero overall spin. However, an unpaired proton and unpaired neutron will have a lower energy when their spins are parallel, not anti-parallel. This parallel spin alignment of distinguishable particles does not violate the Pauli exclusion principle. The lowering of energy for parallel spins has to do with the quark structure of these two nucleons. As a result, the spin ground state for the deuteron (the nucleus of deuterium, the 2H isotope of hydrogen), which has only a proton and a neutron, corresponds to a spin value of 1, not of zero. On the other hand, because of the Pauli exclusion principle, the tritium isotope of hydrogen must have a pair of anti-parallel spin neutrons (of total spin zero for the neutron spin-pair), plus a proton of spin 1/2. Therefore, the tritium total nuclear spin value is again 1/2, just like the simpler, abundant hydrogen isotope, 1H nucleus (the proton).
=== Absorption === 2,6-DCBQ is mainly absorbed via the oral route, typically through the ingestion of contaminated drinking water. Dermal absorption is also considered a viable pathway following aqueous exposure. The compound's low molecular weight and moderate lipophilicity favor absorption across the gastrointestinal tract. However, the high reactivity may limit total systemic bioavailability. The quinone structure reacts rapidly with biological nucleophiles in physiological fluids, leading to localized interactions before the unchanged compound can reach systemic circulation.
Trinity admonishes Neo to run, but he stands his ground, having begun to believe that he may be the One. Neo and Smith draw guns and fire them empty, but are able to effortlessly dodge each other's fire. Neo skillfully engages Smith in hand-to-hand combat, almost seeming to be Smith's equal. In the end, Neo is briefly incapacitated and held by Smith as a subway train approaches, but at the last minute, he is able to get free and backflip up onto the platform, leaving Smith to be run over. However, the agent possesses the body of the conductor and emerges from the train. Neo, realizing that the agents' ability to possess other bodies makes this a fight he cannot win, flees the subway station. Pursued by Smith and his fellow agents, Neo is able to evade them and reach the location of the landline phone, just to be ambushed and fatally shot in the chest by Smith. Trinity, seeing Neo die in the real world while his mind is still in the Matrix, tells his evidently lifeless body that the Oracle had foretold that she would fall in love with the One. When kissed by Trinity, Neo is revived, finally fully emerging as the One. When the agents try to kill him again, Neo simply raises his hand, and the bullets freeze in mid-air, then drop harmlessly to the ground. It is then shown that he is able to perceive, interpret, and alter the computer code of the Matrix. Completely believing in his new-found powers, he effortlessly fends off agent Smith before forcing himself into the agent's body and destroying it from within. The other two agents quickly flee.

It is a laboratory-made peptide that mimics growth hormone-releasing hormone. It prompts the pituitary gland to release growth hormone and has been studied mainly in adults with HIV-associated lipodystrophy.
Administered growth hormone supplies the hormone directly, while this peptide acts upstream by prompting the pituitary to release it. The indirect route preserves pulsatile secretion and some endogenous feedback, which changes the hormone and IGF-1 profile observed after treatment.
Reductions in visceral adipose tissue appear consistently in randomized trials of the approved population. Effects on peripheral fat, cardiovascular outcomes, and use outside that population are less well established.
It is a synthetic peptide analog of human growth hormone-releasing hormone. It is used clinically to reduce excess visceral abdominal fat in adults with HIV-associated lipodystrophy. It works by stimulating pituitary growth hormone release.
