== Biosynthesis == The production of bottromycin by S. bottropensis and S. scabies, as well as the production of a bottromycin analog termed bottromycin D, has been studied. It was independently confirmed in 2012 by multiple groups that bottromycin is produced as a ribosomal peptide natural product that it subsequently post-translationally modified. Before this, it was unclear whether bottromycin was produced by nonribosomal peptide synthetase machinery (NRPS). The presence of amino acids other than the 20 proteinogenic amino acids is often a feature of NRPS products because NRPS machinery can directly incorporate other amino acids, among other chemical building blocks. Ribosomal peptide synthesis, which is the same machinery that produces all proteins found in the cell, is limited to the 20 proteinogenic amino acids. However, bottromycin was found to be a highly modified ribosomal peptide by a combination of genome mining and gene deletion studies. In ribosomal peptide synthesis, the final product results from modifications to a linear peptide starting material translated by the ribosome from an mRNA transcript. In S. scabies the precursor peptide, termed BtmD, is a 44-amino acid peptide. The precursor peptide is termed BmbC in S. bottropensis. The amino acids forming the bottromycin core are residues 2-9 in BtmD: Gly-Pro-Val-Val-Val-Phe-Asp-Cys. In bottromycin D, the sequence is Gly-Pro-Ala-Val-Val-Phe-Asp-Cys, and the precursor peptide is termed BstA. BstA shares high sequence homology with BtmD in the follower peptide region.
The TNXB gene has an associated pseudogene, TNXA. Both TNXA and TNXB genes are located within the RCCX cluster, which consists of a series of modules with genes close to each other: winged helix repair factor 1 (WHR1), complement 4 (C4), steroid 21-hydroxylase (CYP21), and tenascin-X (TNX). In a monomodular structure of the RCCX cluster, all of the genes are functional, i.e. protein-coding, but if there are two or more modules within the cluster, there is only one copy of each functional gene rest being non-coding pseudogenes with the exception of the C4 gene which always has active copies. The presence of the pseudogene is a consequence of MHC class III locus duplication during evolution. Strong 3' homology between TNXB and TNXA can provoke genetic recombination between the two loci, thus leading to the apparition of TNXA/TNXB chimera.
The maximum recommended dose should not exceed three times the original vortioxetine dose. Vortioxetine and its metabolites show no meaningful interactions with a variety of assessed cytochrome P450 enzymes and transporters (e.g., P-glycoprotein), hence vortioxetine is not expected to influence the pharmacokinetics of other medications importantly. == History == 6-HO-DET was first described in the scientific literature by Stephen Szara and colleagues by 1962. It was identified as a major active metabolite of diethyltryptamine (DET). In addition, they found that excretion of 6-HO-DET with DET administration correlated with DET's hallucinogenic effects and that 6-HO-DET was much more potent than DET in humans based on preliminary observations. Consequently, Szara and colleagues theorized that 6-hydroxylation of psychedelic tryptamines like dimethyltryptamine (DMT), DET, and α-methyltryptamine (AMT) was importantly involved in their hallucinogenic effects. However, this hypothesis was later found to be incorrect and was abandoned.
