=== Discovery and development === Aptamers were originally discovered in 1990 when Lary Gold and Craig Tuerk utilized a method of directed evolution known as SELEX to isolate a small single stranded RNA molecule that was capable of binding to T4 bacteriophage DNA polymerase. Additionally, the term “aptamer” was coined by Andrew Ellington, who worked with Jack Szostak to select an RNA aptamer that was capable of tight binding to certain organic dye molecules. The term itself is a conglomeration of the Latin “aptus” or “to fit” and the Greek “meros” or “part." RNA aptamers are not so much “created” as “selected.” To develop an RNA aptamer capable of selective binding to a molecular target, a method known as Systematic Evolution of Ligands by EXponential Enrichment (SELEX) is used to isolate a unique RNA aptamer from a pool of ~10^13 to 10^16 different aptamers, otherwise known as a library. The library of potential aptamer oligonucleotides is then incubated with a non-target species so as to remove aptamers that exhibit non-specific binding. After subsequent removal of the non-specific aptamers, the remaining library members are then exposed to the desired target, which can be a protein, peptide, cell type, or even an organ (in the case of live animal-based SELEX). From there, the RNA aptamers which were bound to the target are transcribed to cDNA which then is amplified through PCR, and the PCR products are then re-transcribed to RNA.

== Plot summary == Anathem is set on the fictional planet of Arbre. Thousands of years before the events in the novel, the planet's intellectuals entered concents (monastic communities) to protect their activities from the collapse of society. The avout (intellectuals separated from Sæcular society) are banned from possessing or operating most advanced technology and are supervised by the Inquisition, which answers to the outside world. The avout are normally allowed to communicate with people outside the walls of the concent only once every year, decade, century, or millennium, depending on the particular vows they have taken. The narrator and protagonist, Fraa Erasmas, is an avout at the Concent of Saunt Edhar. His teacher, Fraa Orolo, discovers that an alien spacecraft is orbiting Arbre – a fact that the world government (Sæcular Power) attempts to cover up. Erasmas becomes aware of Orolo's discovery after Orolo is banished (in a rite called Anathem) from the concent for using a video camera (a forbidden technology) to observe the ship. The presence of the alien ship soon becomes an open secret among many of the avout at Saunt Edhar. The alien ship eventually declares its presence by shining a laser upon several Millenarian Maths (the bastions of those avout who have taken a thousand-year vow of isolation). Shortly after that, the Sæcular Power summons many avout from Saunt Edhar, including Erasmas and a Millenarian named Fraa Jad. The avout are told to travel to the concent of Saunt Tredegarh to attend a Convox (a joint conference of the avout and the Sæcular Power).

=== Jerry Conner === Jerry Garcia Conner is played by Cole Roberts and Morgan Roberts. Jerry is the youngest of Roseanne and Dan's children, born in 1995 when the two are in their forties. Jerry's conception presented a struggle for Roseanne and Dan, who previously never experienced fertility issues. He was named after musician Jerry Garcia of the Grateful Dead, who had a fatal heart attack during Roseanne's pregnancy and of whom she and Dan were fans. The decision to add a pregnancy storyline to the series coincided with Roseanne Barr's real-life pregnancy with her son, Buck Thomas. Barr, like her character, struggled with fertility, though Barr ultimately resorted to fertility drugs to conceive while her character became pregnant naturally. Initially, it was planned that the new cast addition would be a daughter, but when Barr gave birth to a boy, she decided the Conners should have a son instead, wanting her on-screen child's sex to match her newborn child's. Once it was decided the baby would be a boy, Barr named the character after Jerry Garcia and intended to have Roseanne give birth at a Grateful Dead concert. However, plans for a concert birth were scrapped when Garcia died unexpectedly. Because he was so young, Jerry did not play a significant role in the series' initial run.
