Amarasate is a supplement developed to trigger the "Bitter Brake" satiety mechanism, whereby bitter foods targeted to the small intestine trigger the release of the same appetite suppressing gut hormones that are typically elevated after a meal. The Bitter Brake concept was developed based on a combination of historical data supporting the use of bitter foods as a means to suppress appetite, and the discovery of bitter taste receptors (TAS2Rs) in appetite regulating enteroendocrine cells of the gastrointestinal tract. In vitro and biopsy research had identified TAS2Rs throughout the gastrointestinal tract and over 900 plant compounds were tested, with amarasate found to be a potent TAS2R activator. The role of gastrointestinal taste receptors has been examined in animal and human trials and a 2021 meta-analysis determined that pre-meal treatment with bitter compounds reduced energy intake, concluding that "bitter stimuli are most potent to influence eating behaviour".

== Research career and academic posts == After completing graduate school, Fenn's first job was with Monsanto, working in the Phosphate Division and producing polychlorinated biphenyls (PCBs). Fenn and his colleague James Mullen became disenchanted with the direction of work at Monsanto, and they resigned together in 1943. Fenn worked briefly at a small company named Sharples Chemicals that focused on the production of amyl chloride derivatives. In 1945, he joined Mullen at his new startup, Experiment, Inc, focusing on research and development. Fenn's first publication came in 1949 as a result of his work with Mullen. That this publication came ten years after he completed graduate school made Fenn somewhat of a rarity amongst academics. In 1952, Fenn moved to Princeton University as Director of Project SQUID, a program to support research related to jet propulsion that was funded by the Office of Naval Research. During this period, Fenn started his work developing supersonic atomic and molecular beam sources, which are now widely used in chemical physics research. After working with Project SQUID, Fenn returned to Yale University in 1967. He held a joint appointment in the chemistry and engineering departments until 1987, conducting much of his research in Mason Laboratory. In 1987, Fenn had reached Yale's mandatory retirement age. He became a professor emeritus, entitling him to office space at the university, but costing him most of his laboratory space and research assistants.
Eventually the funk project broke up, and in 1987 Staley joined Cantrell's band on a full-time basis. Two weeks after the band's formation, they were playing a gig at Washington State University, trying to fill in a 40-minute set with a couple of original songs along with Hanoi Rocks and David Bowie covers. The band played a couple of gigs in clubs around the Pacific Northwest, calling themselves different monikers, including Diamond Lie, the name of Cantrell's previous band, and "Fuck", before eventually adopting the name that Staley's previous band had initially flirted with, Alice in Chains. Staley contacted his former bandmates and asked for permission to use the name. Nick Pollock was not particularly thrilled about it at the time, and thought he should come up with a different name; both he and James Bergstrom ultimately gave Staley their blessing to use the name.
By 1982, the perceived passivity of the FARC, together with the relative success of the government's efforts against the M-19 and the ELN, enabled the administration of the Liberal Party's Julio César Turbay Ayala (1978–82) to lift a state-of-siege decree that had been in effect, on and off, for most of the previous 30 years. Under the latest such decree, President Turbay had implemented security policies that, though of some military value against the M-19 in particular, were considered highly questionable both inside and outside Colombian circles due to numerous accusations of military human rights abuses against suspects and captured guerrillas. Citizen exhaustion due to the conflict's newfound intensity led to the election of President Belisario Betancur (1982–1986), a Conservative, with 47% of the popular vote. Betancur directed peace feelers at all the insurgents and negotiated a 1984 cease-fire with the FARC at La Uribe, Meta, after a 1982 release of many guerrillas imprisoned during the previous effort to overpower them. A truce was also arranged with the M-19. The ELN, however, rejected any negotiations and continued to rebuild through the use of extortion and threats, in particular against oil companies of European and U.S. origin. At the same time as these developments, the growing illegal drug trade was becoming increasingly important to all participants in the Colombian conflict. Guerrillas and newly wealthy drug lords had mutually uneven relations, and numerous incidents occurred between them.

=== Pharmacodynamics === Isoergine shows affinity for serotonin receptors labeled with serotonin or LSD in rat brain membranes. It has about 10- to 25-fold lower affinity for these receptors than serotonin or LSD and has the same affinity as ergine and iso-LSD (IC50Tooltip half-maximal inhibitory concentration = 100–200 nM for isoergine, 200 nM for ergine and iso-LSD, and 8–10 nM for LSD). No other receptor interaction data are available for isoergine as of 2020. However, computer-predicted receptor affinities are available for ergine/isoergine (stereochemistry not taken into account). The drug is said to have about 4.3% of LSD's antiserotonergic potency in vitro. It is 5- to 33-fold less potent than LSD in producing behavioral changes in the conditioned avoidance test in rodents.

=== Further clinical studies of radiolabeled somatostatin receptor antagonists === Ga-NODAGA-JR11 had entered further clinical studies as an imaging agent, while Lu-DOTA-JR11 had similar research done as a therapeutic agent, as JR11 has a high binding affinity for ssrt2 subtype receptors which are highly expressed on the surface of tumor cells. Gallium-containing agonists had already been established as an imaging agent. Lutetium-containing agonists were used as a therapeutic agent in peptide receptor radionuclide therapy, due to the lower energy electrons emitted, and γ-emission causing easier dose adjustment to patient characteristics to avoid renal damage. The NODAGA chelator was used over DOTA in Gallium antagonists due to higher binding affinity, while no Lu-NODAGA compounds were developed due to established usage of Lu-DOTA derivative agonist drugs, and poor uptake compared to DOTA, which is reverse that of the gallium-containing antagonists.
